The oyster vasa-like gene was previously demonstrated to be specifically expressed in germline cells of adult oysters Crassostrea gigas. In the present study, this gene was used as a molecular marker to establish the developmental pattern of germline cells during oyster ontogenesis. using whole-mount in situ hybridization and real-time PCR. The Oyvlg transcripts appeared to be localized to the vegetal pole of unfertilized oocytes and maternally transmitted to embryos. At early development, these maternal transcripts were observed to segregate into a single blastomere, from the CD macromere of 2-cell stage to the 4d mesentoblast of blastula. From late blastula stage, the mesentoblast divided into two cell clumps that migrated to both sides of the larvae... |

During a series of experiments, bacteriological elements in scallop larval rearing were investigated: larvae susceptibility to pathogens as a function of their age, and the use of probiotic bacteria during larviculture. Younger larvae (d5 PF) were highly more susceptible to pathogenic-challenge than their older siblings, which were challenged at an older age (d15 PF). A challenge with 104 CFU mL−1 of V. pectenicida killed 100% of d5 PF larvae 7 days following challenge, yet killed only 9% of d15 PF larvae 9 days following challenge. Use of the probiotics Phaeobacter gallaeciensis, Alteromonas macleodii 0444 and Neptunomonas sp. 0536, provided for larger larvae, a high yield of competent larvae and, perhaps more importantly, protection against... |

The haptophyte Pavlova lutheri was cultivated in a 4-l flat alveolar photobioreactor (FAP) in semi-continuous mode during 35 days, achieving a mean volumetric productivity of 0.4 g l-1 day-1. The microalgal culture was harvested from the photobioreactor at an average concentration of 3.5 g l-1 and subjected to different storage conditions in which the effects of temperature (1 °C, 4 °C and 25 °C), air supply and preservative (ascorbic or propionic acid) addition on the viability of the stored suspensions were investigated. Low temperatures (1 and 4 °C) and air-bubbling extended the shelf-life of the stored suspension, while no beneficial effects of ascorbic or propionic acid were found. The influence of temperature and air-bubbling on fatty acid content... |

A novel technique was developed for the flocculation of marine microalgae commonly used in aquaculture. The process entailed an adjustment of pH of culture to between 10 and 10.6 using NaOH, followed by addition of a non-ionic polymer Magnafloc LT-25 to a final concentration of 0.5 mg L−1. The ensuing flocculate was harvested, and neutralised giving a final concentration factor of between 200- and 800-fold. This process was successfully applied to harvest cells of Chaetoceros calcitrans, C. muelleri, Thalassiosira pseudonana, Attheya septentrionalis, Nitzschia closterium, Skeletonema sp., Tetraselmis suecica and Rhodomonas salina, with efficiencies ≥80%. The process was rapid, simple and inexpensive, and relatively cost neutral with... |

Pathogenic bacteria are known to be one of the main factors affecting Pecten maximus larval survival in hatcheries. As a result, juvenile production often relies on the use of antibiotics during larval culture. However, limitations of the usage of chemicals such as chloramphenicol in aquaculture have been reinforced due to their negative environmental impact and alternatives are accordingly needed. Thus, the importance of bacterial transfer from oocytes to larvae has been studied here as well as procedures to limit larval mortality in P. maximus. In order to reduce bacterial contamination during larval development, we focused on two periods, broodstock conditioning and post-fertilization. The animals were conditioned for 2 months with two erythromycin... |

The 2010 Deepwater Horizon (DWH) oil spill released millions of barrels of oil and dispersant into the Gulf of Mexico. The timing of the spill coincided with the spawning season of Crassostrea virginica. Consequently, gametes released in the water were likely exposed to oil and dispersant. This study aimed to (i) evaluate the cellular effects of acute exposure of spermatozoa and oocytes to surface slick oil, dispersed mechanically (HEWAF) and chemically (CEWAF), using flow-cytometric (FCM) analyses, and (ii) determine whether the observed cellular effects relate to impairments of fertilization and embryogenesis of gametes exposed to the same concentrations of CEWAF and HEWAF. Following a 30-min exposure, the number of spermatozoa and their viability were... |

In mollusc hatcheries and nurseries, live microalgae are traditionally used as feed for bivalves. Despite recent improvements in efficiency, such intensive culture of microalgae represents major cost factors in the production of bivalve larvae and spat. In an attempt to provide cost-effective alternatives and to simplify hatchery-nursery procedures, non-living algal food has been developed over the past few years. Substitution products such as bacteria, yeast, vitamin or docosahexaenoic acid enriched powders, dry and concentrated microalgae and microcapsules have been tested and evaluated on an experimental scale. The present article reviews major research aimed at developing non living diets in mariculture, which generally resulted in lower growth and... |

Since 2008, mass mortalities of 1-yr-old Crassostrea gigas associated with the ostreid herpesvirus OsHV-1 μVar have occurred along all the coasts of France when seawater temperature reaches 16 to 17°C. The present study aimed to characterize the effect of temperature on oyster survival in combination with OsHV-1 DNA quantification by standard real-time PCR and total vibrio population levels in oyster tissues. To examine the effect of seawater temperature on disease transmission and related mortality of oysters, cohabitation experiments were conducted between healthy naïve oysters and oysters previously exposed to field conditions in areas where mortalities were occurring. Oysters initially maintained in controlled conditions (free of mortality and negative... |

Because of diseases affecting fIat oyster Ostrea edulis, acclimatization of the Argentina oyster Ostrea puelchana has been tested in 1989-90 in five French oyster farming sites. Following ICES recommendations , hatchery produced juveniles, from parents imported from Argentina, were transferred to experimental si tes in summer 1989. Very high mortalities occurred, reaching 46 to 98% in March-Apri1 1990. They continued later on, resulting in the end of experiments in March 1991. The growth of surviving oysters was relatively poor in comparison of the local species. Parasites Marteilia refringens and Bonamia ostreae have been detected but at rates inadequate ta explain the high mortalities. AlI these results lead not to retain this species for breeding on... |

The lipid class and the fatty acid compositions of microalgae highly influence bivalve larval and post-larval development. Light is an essential environmental factor for microalgal culture, and quantity and quality of light may induce changes in the biochemical composition of the algae. The objective of this study was to investigate the effect of light spectrum (blue vs. white light) on lipid class and fatty acid compositions of Tisochrysis lutea cultured in a chemostat. Two different dilution rates (D) were assayed for each light spectrum: 0.2 and 0.7 day−1. Triacylglycerol (TAG), sterol, and hydrocarbon (HC) content increased sharply at low D. The proportion of alkenones was significantly reduced under blue light. Polyunsaturated fatty acids (PUFA), and... |

To determine the effects of food quality on Ostrea edulis reproduction, European flat oysters were conditioned during two sets of experiments, carried out in spring and autumn, during 40 days at 19 degrees C, in 50 l transparent flow-through tanks, in triplicate, and fed constantly at 900 mu m(3) mu l(-1), with eight different types of microalgae. Four species were fed per group of trials: Isochrysis affinis galbana, Chaetoceros gracilis, Skeletonema marinoi, and Tetraselmis suecica were fed to flat oysters in the first set; whereas Rhodomonas salina, Thalassiosira weissflogii, Thalassiosira pseudonana, and Pavlova lutheri were provided during the second set of experiments. At the beginning and end of both conditioning periods, oysters were sampled and... |

In an attempt to provide cost-effective alternatives to live microalgae and to simplify hatchery-nursery procedures, concentrated microalgae appear to be very promising. Due to its high preservation ability, Prasinophytes appear to be good candidates for such alternative diets. Nevertheless, in a previous study the utilization of Tetraselmis suecica was only conceivable for old Crassostrea gigas larvae. In the present work its nutritional quality has been accordingly searched on ]apanese oyster's juveniles. Whatever the assemblages and condition, fresh or preserved microalgae, T. suecica did not allow an adequate postlarval development in C. gigas. |

Pecten maximus larvae exhibit amylase as a main digestive enzyme indicating the possibility of starch hydrolysis. The food value of Rhodomonas salina and Porphyridium cruentum, both rich in starch, on the development of Pecten maximus larvae was investigated. The larval development was studied by means of an image analysis technique and epifluorescence microscopy was used for detecting ingestion and digestion of each phytoplankton species. The biochemical contents of algae and larvae were also assessed. The best growth of Pecten maximus larvae was observed when fed with the mixture PTS (plurispecific control). No growth was observed for larvae fed with Porphyridium cruentum of which most of the population died on day 16 (90%). This algal species was... |

Four microalgae species (Rhodomonas salina, Thalassiosira weissflogii, Thalassiosira pseudonana and Pavlova lutheri) were evaluated to estimate their potential as food for Ostrea edulis (L.) reproductive conditioning. Best ingestion and absorption were observed with R. salina (3.44 and 1.59 mg g− 1 h− 1, respectively), followed by T. pseudonana (2.75 and 0.98 mg g− 1 h− 1) and P. lutheri (2.40 and 0.91 mg g− 1 h− 1). Oysters fed T. weissflogii exhibited the lowest ingestion and absorption values (1.40 and 0.68 mg g− 1 h− 1). Proximate composition (proteins and carbohydrates) and lipid content (fatty acids and sterols) analysed in four main tissues (gonad, digestive gland, muscle and gills) also differed significantly with diet. Protein ranged from 355 mg... |
